Prof. Fernandez et al. report on a five year long study of characterization and identification of anti-malarials in Africa using DESI and DART mass spectrometry. DESI is utilized in a 'reactive' mode to detect intact artemenisins, whereas in conventional ESI or DART the compounds fragment that results in less confident identification.

DESI, MALDI and Histochemistry on the same sample

Eberlin et al. have developed methods for performing DESI, MALDI and H&E staining on the same tissue sample. The workflow begins with DESI imaging using non-destructive solvents, then matrix application and MALDI analysis, followed by H&E staining. While this new procedure was implemented on different instruments in separate laboratories, the concept can be advantageously applied in dual source systems such as Waters Synapt G2 or the Bruker SolariX.

Prof. Fernandez' most notable work is in the detection of counterfeit antimalarial tablets, which will be discussed here among other examples.

Abstract

Although more rampant in the developing world, counterfeit drugs know no borders, and cases are constantly uncovered in both developed and developing countries. For example, in June 2010 the US Food and Drug Administration issued a warning of online pharmacies selling a fake version of the flu treatment Tamiflu (oseltamivir) that could be dangerous to people allergic to penicillin. The so called "generic Tamiflu" sold online was found to contain cloxacillin, a penicillin that could cause unexpected and inexplicable severe allergic reactions. In January 2010, INTERPOL released the results of a large multi-country police operation targeting the manufacture and distribution of counterfeit medicines in SE Asia named "STORM II". This operation resulted in more than 30 arrests and the seizure of 20 million fake and illegal medicines. It also led to the closure of more than 100 pharmacies and illicit drug outlets. In this webinar, we will showcase the use of Desorption Electrospray Ionization-Mass Spectrometry (DESI-MS) for the qualitative and quantitative analysis of solid pharmaceutical samples, with specific emphasis on how "unknown knowns" can be identified, how active ingredients can be quantified with or without the use of internal standards, and how DESI imaging MS can be used for investigating product homogeneity.

Profiling Triglycerides Directly from Food Products using DESI-MS

In combination with a Waters LCT Premier, DESI was used to profile triglycerides on the surfaces of food products. Tostitos chips were ground and extracted with MTBE. The extract was spotted on Whatman paper and analyzed using a 70/30 methanol and 10 mM ammonium acetate as the spray solvent. Five lipid species were identified using exact mass measurements as ammonium adducts. A Tostitos chip was then analyzed directly using 3/1/ 0.003% methanol / chloroform / ammonium acetate spray solvent. Remarkably, the triglyceride profiles from the extracted sample and the sample analyzed directly using DESI produced the same results.

Abstract

The detection and identification of protein residues from archaeological artefacts has been the subject of considerable controversy over the past two decades, this controversy spurring the application of a range of different analytical methods. Part of the debate surrounds the ability of the methods to uniquely identify original proteins. Another is the apparent contradiction between the ease of extraction in the laboratory of residues that have managed to survive in sediment over millennia; some studies use simple solvents including water, whilst others claim that efficient extraction requires the destruction of the artefact.
Desorption electrospray ionisation mass spectrometry (DESI-MS) offers the potential to directly analyse proteins bound to the surface of artefacts. The method permits desorption of analytes under ambient conditions directly from a wide range of surfaces with little or no sample preparation. This has already led to its application in a range of areas, including forensic analysis of trace levels of narcotic drugs. It does not require destruction of the artefact (e.g. by drilling or dissolution) and can work in concert with other methods (for instance it could be used as a screening tool prior to lipid extraction).
Here we report the construction of a DESI-MS interface and consider the potential of the method to contribute to the debate surrounding the preservation of proteins on artefacts. The DESI-MS interface was fitted to a conventional ion trap mass spectrometer and tested using peptides and proteins artificially applied to the surface of flint flakes and potsherd samples. A range of intact proteins was analysed; the DESI interface identified test proteins up to a maximum molecular weight of approximately 20 kDa. Peptides generated from these and larger proteins could be detected using DESI-MS following tryptic digestion in situ. The collected data allowed identification of the originating proteins. The potential applications to archaeological science of this new analytical tool are discussed.

DESI is used for rapid screening of anabolic steroids in hair

Dr. Nielen et al. have shown the feasibility for examining hair samples for anabolic steroids using DESI-MSn. They combined the Omni Spray 1-D source with a Thermo Scientific LXQ linear ion trap mass spectrometer to record both full scan and data dependent MSn spectra of hair samples. The authors conclude that DESI-MSn analysis could be suitable for rapid screening for prioritizing samples for follow-up investigation.

Direct analysis of camptothecin from Nothapodytes nimmoniana by desorption electrospray ionization mass spectrometry (DESI-MS)

Prof. T. Pradeep et al. describe the use of the Omni Spray 2-D ion source for fast and direct ambient detection of the anti-tumor drug, camptothecin, and its derivative, 9-methoxycamptothecin in Nothapodytes nimmoniana. Different parts of the plant such as leaves, stems and bark were examined. The ion intensities suggested that the concentration in bark is higher than that in the leaves and stems. The method does not require any sample preparation or preseparation.

Summary:

By integrating a 3-dimensional navigation system with an ultrasonic surgical probe, the authors obtained image-registered surgical specimens and subjected those specimens to intraoperative molecular analysis analysis by DESI-MS and MS/MS.The mass spectral results were then validated against standard histopathology. The results of this proof of concept study showed correlation between lipid constitution of the tumor mass and malignancy.

New Publication: Elution, Partial Separation, and Identification of Lipids Directly from Tissue Slices on Planar Chromatography Media by Desorption Electrospray Ionization Mass Spectrometry

A method for the examination of intact tissue sections for gangliosides and other lipids using desorption electrospray ionization (DESI) mass spectrometry (MS) is presented. In the present work, thin tissue slices (16 μm) taken from the rat brain are thaw mounted onto planar chromatographic media and the lipids are eluted, partially separated, and analyzed directly from the plate by DESI-MS in the negative ion mode. With the lanes scanned parallel to the direction of the chromatographic separation in the full scan mode, the selected ion current associated with ions of separated lipid molecules is plotted in order of increasing Rf values. Distinctly different classes of lipids are detected using this method, including several ganglioside species (i.e., GQ1, GT1, GD1, and GM1) and sulfoglycosphingolipids. For the examination of gangliosides in the full scan negative ion mode from high-performance thin-layer chromatography (HPTLC) plates, the limit of detection (LOD) was determined to be approximately 3 pmol. Tandem mass spectrometry (MS/MS) using the linear ion trap was used to confirm the presence of selected gangliosides and other lipids directly from the HPTLC plate. DESI-MS/MS revealed the presence of both the GD1a and GD1b isomers. The simplicity of this approach where planar separations are relied upon for sample preparation and presentation to the MS should allow for the examination of a variety of complex samples including the rapid examination of foodstuffs, bacteria, whole blood, and needle biopsies for cancer diagnostics.
